Singampuli made his acting debut in the Tamil film ‘Nan Kadavul’ in 2009.
He has acted in Muthuku Muthaka (2011), Muthina Kathirika (2016), Kaviya Thalaivan (2014), Thavam (2019), Nanga Rompa Pisi (2020) and many other Tamil films.
He made his directorial debut in 2002 with the Tamil film ‘Red’. He also acted in the film ‘Mayavi’ (2005).
In 2010, Singampuli won the Ananda Vikatan Cinema Award for Best Comedian for the film Mayandi Kudumbathar (2009).

- In an interview, Singampuli said that he watches 400 films a year. He also said that he has been interested in acting since childhood.
- After graduating from college in Bangalore, he came to Chennai to look for a job in cinema. Initially worked as an assistant director.
- He also worked as a dubbing artist in the role of Timon in the 2019 film The Lion King.
